In a trans-sialylation reaction facilitated by Tr15, sialic acid from CGMP is moved using the whey-derived LacNAc to produce 3'SLN. This is a useful and cost-effective manufacturing method. In addition, natural resources can be used to produce 3'SLN.
Five monosaccharides serve as the building blocks for the neutral and anionic species that make up human milk oligosaccharides (HMOs). These constituents include L-fructose (Fuc), D-glucose (Glc), D-galactose (Gal), N-acetylglucosamine (GlcNAc), and N-acetylneuraminic acid (or sialic acid; NeuAc). Sialic acid, one of the specific components found in HMOs, is essential for the growth of neurons, the development of the brain, neuronal transmission, cognition, and synaptogenesis.

A class of glycosyltransferases known as sialyltransferases generates sialylated glycans in mammalian systems. Due to the absence of organelle membrane structures, it is notoriously challenging to produce mammalian sialyltransferases in Escherichia coli systems. Therefore, LacNAc was originally generated and purified directly from whey in the majority of current research. The sialic acid from CGMP was then transferred using the whey-derived LacNAc in a trans-sialylation process catalyzed by Tr15 to create 3'SLN. This is a practical and affordable production technique. Additionally, 3'SLN can be obtained from natural sources, but there is no acceptable method for meeting the need on a big scale.
