Galactomannan produced by the human fungal pathogen A. fumigatus is an iconic biomarker including mannose and galactofuranose. Galactomannan produced by A. fumigatus is a unique fungal polysaccharide that contains three forms: membrane-bound via GPI anchors, cell wall-localized galactomannan, and free galactomannan. The biosynthetic pathway of galactomannan in A. fumigatus requires four steps: (1) Synthesis and transport of nucleotide sugars; (2) Polymerization of galactomannan through glycosyltransferase; (3) Transport of lipo-galactomannan to the surface of the plasma membrane within the cell; (4) Cross-link galactomannan to β-1,3-glucan in the cell wall. In addition, galactomannan is one of the key antigenic molecules of A. fumigatus, which is of great significance for the diagnosis of invasive bronchial disease.
