Bioactivity of Acetyl Coenzyme A (Ac-CoA)

Ac-CoA is a membrane-impervious central metabolic intermediate, the actual form for pyruvate entering the tricarboxylic acid cycle (TCA cycle) during glycolysis, a vital precursor of lipid synthesis, the unique donor of acetyl acetylation, and an effective allosteric activator of pyruvate carboxylase. In addition, it is also a source of the neurotransmitter acetylcholine. In short, Ac-CoA has indispensable importance in living organisms.

Synthetic Strategies

Ac-CoA can be conventionally obtained by chemical synthesis using acetic anhydride and CoA as raw materials. Enzyme catalysis strategy and biosynthesis strategy are used more frequently. Ac-CoA can be synthesized from 1-acetylimidazole and CoA by the metabolism of Bacillus subtilis. It can be obtained from sodium pyruvate and CoA by the action of bacterial pyruvate dehydrogenase complex. If Acetoacetyl-CoA is available, it is a very rapid method to break down acetoacetyl CoA into Ac-CoA by thiolysis. In the presence of acetate CoA-transferase, Ac-CoA can be converted from the reaction of acetic acid and succinyl-CoA.

  • Ac-CoA in assays of citrate synthase activity: In the TCA cycle, citrate is formed by the condensation of oxaloacetic acid and Ac-CoA catalyzed by citrate synthase. Ac-CoA can be used as a substrate and energy donor to test citrate synthase activity.
