Introduction of 3'SLNB Production Methods

The unique human milk oligosaccharide (HMO) blueprint pattern is made up of (up to) five distinct monosaccharides: D-galactose (Gal), D-glucose (Glc), D-N-acetylglucosamine (GlcNAc), D-L-fucose (Fuc), and the sialic acid D-N-acetylneuraminic acid (Sia). The overall diversity of HMO structures consists of five distinct monosaccharides and ten different kinds of connections. With the help of precise blueprint patterns, it is now feasible to synthesize a variety of HMO structures in vitro or in vivo by concentrating on a small number of distinct enzyme activities.

A significant disaccharide known as lacto-N-biose I has been found to have a significant role in the carbohydrate structure of several glycoconjugates, including tumor-associated antigens. Potential tumor markers have been detected for antibodies to lacto-N-biose I. This moiety is the main part of the Lewisa antigenic determinant. Lacto-N-biose I is 2-3-Sialylated to produce the trisaccharide 3′-sialyl-lacto-N-biose I, also known as 2,3-Sialyllacto-N-biose (3'SLNB), which has been demonstrated to be the fundamental building block of the sialyl-Lewisa, which has been discovered as a functional ligand for E-selectin. During both healthy and pathological inflammatory reactions to injury and infection, it facilitates the trafficking and recruitment of blood-borne leukocytes to endothelial cells.
