ACL is extracted and purified from the A. caudatus plant, which is a dimeric glycoprotein composed of two subunits with a molecular weight between 60,000 - 70,000 and binds best under slightly acidic conditions. The isoelectric focusing of the lectin resolves it into four bands with isoelectric points between pH 6.7 and pH 7.7. We use further chromatography techniques (such as ion exchange chromatography, gel filtration chromatography, etc.) to purify the enriched lectin, remove other impurities, and obtain high-purity ACL.
ACL is a protein with lectin activity that specifically binds to Galβ(1-3)GalNAc (T antigen). We perform activity assays on purified ACL samples using appropriate experimental methods and kits. Typically, the activity of a lectin is assessed by the agglutination of red blood cells caused by its binding to specific sugar residues. ACL is not specific for blood types, but has a higher affinity for type O red blood cells than type B and type A red blood cells.
We provide different specifications and scale ACL production services. We aliquot the purified ACL into appropriate containers and store it as required to ensure its stability and shelf life. Results: The authors used lectin microarrays to analyze endometrial cancer tissue and cell lines from patients and divided patients into different clinical stages based on cancer progression (Figure 2A). The results showed that there were significant differences in the relative binding strength of ACL to endometrial cancer in different stages, with the ACL signal in stage III/IV tissue being higher than that in stage I tissue (Figure 2B). The authors believed that the strong correlation between lectins and endometrial cancer stage provided a new way to determine treatment options and predict prognosis.
