Bauhinia purpurea Lectin (BPL) Production Services at CD BioGlyco

Lectins are a class of non-enzymatic, non-antibody glycan-binding proteins that are widely distributed in animals, plants, and microorganisms.   • Extraction and purification
    • First, we grind B. purpurea seeds and use saline extraction. Then, the supernatant is concentrated and then subjected to salting out. Next, the precipitate is taken and desalted using a dialysis operation. Finally, lyophilization is performed to obtain the crude lectin.
    • We acidify the crude lectin product. The pH is adjusted by adding a buffer to it, followed by column chromatography to wash out the heterogeneous proteins and collect the target product. Finally, pure BPL is obtained by dialysis to remove glycans and lyophilization.
  • Performance characterization
    • Molecular weight and subunit molecular weight
      The molecular weight of lectin is determined by gel permeation chromatography, and the molecular weight of the subunit is determined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis.
    • Structure
      The primary, secondary, and tertiary structures of BPL are determined using circular dichroism.
    • Isoelectric point
      The isoelectric point of BPL is obtained by measuring the pH of each part in an amphoteric electrolyte carrier with an acidimeter and comparing it with the stained protein bands.
    • Sugar content
      Glucose is used as the standard sugar and measured by anthrone colorimetry.
    • Agglutination activity
      After doubling dilution on the coagulation plate, add a small amount of glycan solution to each hole, then add erythrocyte suspension, mix and let it stand, and observe the agglutination activity using a microscope.
    • Stability
      Thermal stability: Sampling at different temperatures to check the agglutination activity.
      Acid-base stability: The samples are kept warm in different buffers and then the agglutination activity is determined.

In addition, we will determine the sugar-binding specificity of BPL. It is found to have a binding affinity for d-galactose and d-glucose patterns.

Results: By X-ray crystallography, the tertiary structure of Bauhinia forficata lectin (BFL) was determined to generate the structure of hemagglutinin alone and bind to different ligands. The results indicated that BFL was formed by the interaction of two large antiparallel β-sheets, as shown in Fig.2. However, no α-helix was reported in its structure, consistent with the secondary structure, which contained 19% of the α-helix of BFL. In addition carbohydrate binding was mainly formed by four discontinuous segments of the lectin and the presence of different ligands did not produce any significant structural changes, suggesting a high degree of structural conservation of the ligand-binding pocket.
