Introduction

Carbohydrates as structural materials and energy sources are probably one of the most abundant and challenging biomolecules. They exist either as a single molecule or in combination with other molecules. For example, they bind to proteins to form glycoproteins and covalently link to lipids to form glycolipids. Carbohydrates play important roles in living organisms as important nutrients and major energy sources. Meanwhile, carbohydrates are also involved in a variety of immune responses. Glycans are complex molecules that exist on the cell surface and are involved in various biochemical reactions such as cell recognition and cell communication.

Carbohydrate Separation, Purification and Analysis

Carbohydrates are difficult to obtain in pure form due to the heterogeneity of glycans. Unlike peptides, the spatial orientation of the atoms of carbohydrates determines their properties. Furthermore, the polar, hydrophilic, and highly branched properties of carbohydrates complicate the development of general methods for the separation and purification of carbohydrates. Currently, ion-exchange chromatography (IEC) combined with electrochemistry is widely used for the analysis of carbohydrates, but this method suffers from low capacity and incompatibility with mass spectrometry (MS). Therefore, the development of methods for the separation, purification, and structural analysis of carbohydrates in complex matrices is highly desirable.
