The pathogenic mechanism of many diseases lies in changes in the quantity and/or quality of complex carbohydrates. Mass spectrometry (MS) methods are very promising in monitoring and detecting these significant biological changes. The isotopic labeling method is helpful to accurately quantify the level of released glycans. Quantitative derivatization of free reducing glycans through a variety of methods is the basis of differential isotope labeling of glycans. Isotopic labeling strategies include derivatization of glycan main chain by permethylation with reagents containing heavy or light isotopes, or reductive amination of the reducing end of glycans with light or heavy isotope labeling reagents.
Glycan reductive isotope labeling has been reported as a new glycomics method. Free glycans can be derived by reductive amination of stable isotope labels [12C6]-aniline and [13C6]-aniline with differential coding. And two samples were labeled as light and heavy, respectively, which were recovered by reverse phase chromatography and quantified according to UV-absorbance and relative ion abundance. This technology provides necessary advances in the field of glycomics.
