- State
Chlorhexidine gluconate is a clear, colorless slight yellow, and slightly viscous liquid. It is miscible with water and soluble in ethanol or acetone.
- Identification
- We dilute the sample well with water and copper sulfate solution, then, the solution is boiled to cause the precipitate to coalesce. If a lavender color appears in the solution, the sample is chlorhexidine gluconate.
- Add ferric chloride solution to an aqueous solution of the sample and heat to boiling. If the solution changes color to yellow, the sample is chlorhexidine gluconate.
- The sample is added to a hot solution of cetyltrimethylammonium bromide to which a bromine test solution is added along with a sodium hydroxide test solution. If the solution shows a deep red color, the sample is chlorhexidine gluconate.
- Acidity
The acidity of chlorhexidine gluconate is tested using a pH meter.
- Detection of p-chloroaniline
We dilute chlorhexidine gluconate well with water, hydrochloric acid, and ethanol and compare the color of the solution with that of a p-chloroaniline solution against a black background. If the colors are identical, p-chloroaniline is present in chlorhexidine gluconate.
- Impurity assay
A sample of chlorhexidine gluconate is added to the acetic acid solution. Next, the solution is unfolded on the same thin-layer silica gel plate as the solution of chlorhexidine gluconate standard according to wave-layer chromatography. Finally, the thin layer plate is placed under an ultraviolet light for review. If the sample contains stray dots it means that the sample contains impurities.
- Content assay
Chlorhexidine gluconate is weighed accurately and diluted with water. Then, we add ethanol solution to it. The absorbance is measured spectrophotometrically. Finally, we calculate the content of chlorhexidine from the absorbance value.

Results: In this article, the authors researched the in vivo bioactivity of chlorhexidine gluconate to understand the time required for chlorhexidine gluconate to form a film, the appearance of the film, and its adhesion to the skin. The authors also researched the antimicrobial activity of chlorhexidine gluconate in vitro and in vivo and evaluated optimized formulations for cytotoxicity and wound healing in vitro cell line evaluations. The results showed that transparent homogeneous solutions of chlorhexidine gluconate yielded transparent and pliable films upon actuation and were effective in inhibiting the growth of Staphylococcus aureus in vitro.
