What Is Glycosylation?

Glycosylation is the process of adding carbohydrates to proteins or lipids under the control of enzymes in the endoplasmic reticulum (ER) and Golgi apparatus and is one of the most common post-translational modifications. Proteins are glycosylated to form glycoproteins, which are involved in almost all innate and adaptive immune responses. Protein glycosylation mainly includes the addition of N-glycans, O-glycans, glycosylphosphatidylinositol (GPI) anchors, and phosphorylated glycans to the peptide backbones. Sugars are added to lipids to form glycolipids, such as glycosphingolipids (GSLs). GSLs are among the most abundant glycolipids in humans, performing key cellular functions associated with lipid rafts, and their glycan composition confers specific GSL properties.

Glycosylation in Disease

Glycosylation patterns are one of the first biomarkers of cancer to identify cells with a stem cell-like phenotype in cancer. Glycosylation of specific proteins in serum and tumors is used to assess patient efficacy and prognostic response, such as MUC1 and prostate-specific antigen (PSA). Changes in fucosylation, sialylation, and galactosylation may contribute to autoimmune disease and chronic inflammation. Currently, developments in the field of glycobiology have enabled the development of various glycan-based therapeutics. For example, increased Fc galactosylation in rituximab enhanced complement-dependent cytotoxicity. Therefore, glycosylation services have become an essential part of glycobiology.
