GD1 belongs to the group of gangliosides within sphingolipids. Their structure consists of a ceramide backbone linked to an oligosaccharide unit consisting of six sugar molecules. Two of them are sialic acid. Some GD1 lipids have anti-inflammatory properties by inhibiting the action of bacterial lipopolysaccharides and preventing cytokine activation. In mice, GD1 gangliosides are important for oocyte maturation and fertilization.

CD BioGlyco provides high-quality Disialoganglioside (GD) Analysis Service. We offer a practical protocol for the extraction of GD from biological tissues and liquid chromatography (LC)- tandem mass spectrometry (MS/MS) analysis of the extracted GD. The characterization of the lipid forms of various glycosphingolipids does not depend on their LC profiles, which can avoid uncertainties caused by changes in LC conditions and thus be more reliable and generally applicable. The analysis process mainly includes GD extraction and LC-MS analysis.
Combine the GD standard mixture with a mixture of water, methanol, and chloroform and vortex. After several extractions, the combined upper layers are concentrated on a concentrator and the residue is reconstituted in methanol containing 1% chloroform for LC-MS/MS analysis.
Each GD sample from plasma is injected into a system equipped with a C18 column and pre-column. During the analysis, the autosampler and the column are kept at different temperatures. Elution is carried out by the best program. Acquiring MS or MS/MS spectra using a mass spectrometer, utilizing electrospray ionization (ESI) operating in positive mode. The instrument is programmed for data-dependent acquisition (DDA) of CID using a nitrogen collision gas partner.
In addition, we also provide separation services for GD1 isomers. Matrix-assisted laser desorption/ionization imaging mass spectrometry (MALDI IMS) enables untargeted spatial analysis of gangliosides as well as other biomolecules, directly from tissue sections. Combining capture ion mobility mass spectrometry (TIMS) with MALDI IMS allows in situ gas phase separation of the GD1 isomers GD1a and GD1b.
