Dolichos bifloru Lectin (DBA) Production Service at CD BioGlyco

DBA is extracted and purified from the seeds of the leguminous plant D. biflorus. DBA is a heterotetrameric glycoprotein composed of four subunits of equal size. It has binding activity and is used as a marker for primordial germ cells and spermatogonia to indicate their differentiation status. It also determines the secretion status of individuals with type A blood through hemagglutination inhibition and is used to detect blood types.   • Plant extracts

D. biflorus seeds contain DBA, a phytohemagglutinin that is a natural protein that binds to specific sugar molecules, which has carbohydrate specificity for β-N-acetylgalactosamine.

  • We grind mature D. biflorus seeds to increase cell wall disruption and release of soluble proteins.
  • We use an appropriate buffer to extract the seed powder, dissolve DBA into the extraction solution, and separate the particles through centrifugation and other operations.
  • We purify the extracted mixture through column chromatography or other separation techniques, including ion exchange chromatography, affinity chromatography, etc. By choosing an appropriate purification method, we separate the target lectin from other proteins and impurities.
  • We wash the purified lectins to remove residual impurities and buffers. Appropriate concentration techniques are then used to increase the concentration of the lectin to the desired level.
  • We perform protein identification (such as SDS-PAGE gel electrophoresis) on the produced DBA and conduct activity tests to determine its agglutination ability. Activity testing typically involves experiments with binding to specific glycogen-specific antigens.
  • We will package the purified and characterized DBA appropriately, such as lyophilization or cryopreservation, which ensures the stability and activity of the product during storage.

Results: The authors screened several markers to differentiate and study spermatogonia populations in situ and in vitro using immunohistochemical staining. The results showed that undifferentiated spermatogonia were not labeled by DBA (Fig. 2A), DBA staining intensity (brown precipitate), and labeled cell area increased in pubertal tissue (Fig. 2B), reaching a maximum in active seminiferous tubules (Fig. 2C). This demonstrated that DBA lectin was used to label and isolate undifferentiated spermatogonia from a variety of animal species.

Applications
