- Synthesis of oligosaccharides
We provide one-pot glycosylation to obtain preliminary several oligosaccharides. In this process, we utilize oligosaccharides with special moieties (like oligo-sulfide, and oligo-bromo) as glycosylation donors to induce favorable stereoselectivity and yields in the glycosylation process.
- Synthesis of core oligosaccharides
We use enzymatic reactions to synthesize core oligosaccharides. Based on the oligosaccharides synthesized in the first step, we use glycosyltransferases to construct specific glycosidic bonds and prepare trisaccharides, tetrasaccharides, and so on. In this process, core oligosaccharides with different backbone structures are synthesized using the substrate specificity of glycosyltransferases. At the same time, we provide nuclear magnetic resonance (NMR) technology for structural analysis of the oligosaccharides.
- Construction of oligosaccharide libraries
We utilize enzymatic extension methods to extend each core structure by a different glycosyltransferase (rearrangement of enzyme-modified sequences, etc.). Each extension scheme is designed according to the substrate specificity of the glycosyltransferase to avoid undesirable glycosylation.
In addition, we provide a variety of glycosyltransferase systems to obtain more complex oligos and to avoid creating more prolonged purification steps.
Finally, we provide high-resolution anion-exchange chromatography, electrospray ionization mass spectrometry (ESI-MS), and NMR techniques to characterize the purity and structure of the oligosaccharide library.

Results: The paper describes the process by which a library of 15 different chondroitin sulfate (CS) oligosaccharides is synthesized using enzymes as an alternative to CS. The library includes 4-O sulfate and 6-O sulfate oligosaccharides ranging in size from trisaccharides to non-monosaccharides. The synthesis of each CS oligosaccharide requires additional CS glycosyltransferases, and the high expression of CS glycosyltransferases expands the ability to synthesize a greater diversity of CS oligosaccharides. The authors also demonstrated the synthesis of unnatural 6-O-sulfonated CS pentasaccharides containing 6-O-sulfo-2-azide galactosamine or 6-O-sulfogalactosamine residues. Furthermore, enzymatic synthesis of CS is not considered to allow the preparation of sufficiently large, structurally defined CS oligosaccharides for biological evaluation. Instead, the authors overcame this limitation by increasing the accessibility of recombinant CS biosynthetic enzymes and reducing the production cost of UDP-GalNAc.
