α-Glucosidase from Bacillus stearothermophilus, an exoenzyme, is responsible for hydrolyzing (1→4)-linked α-D-glucose residues with the release of D-glucose.

One unit of α-D-glucosidase activity is defined as the amount of enzyme required to release one µmole of p-nitrophenol per minute from 4-nitrophenyl α-D-glucopyranoside (5 mM), in sodium phosphate buffer (100 mM), pH 6.5 at 40°C.
