Precision Glycan Elongation/branching Pathway Engineering for Enhanced Cell Functionality

The glycan array is capable of displaying oligosaccharides and is also able to report glycan half-antigen binding epitopes as well as predict the structural features of the glycans involved. Therefore, it has an important fundamental role in the biological research of glycans. At CD BioGlyco, we have carried out comprehensive research on Cell-based Glycan Display Arrays and proposed a Glyco-engineered Cell Construction service. Among them, the glyco-engineered cell construction based on the glycan elongation/branching pathway is an indispensable part.

  • Glycation strategy
    To better construct cell lines, we base the delineation of the glycosylation process on glycosyltransferases. Due to the extensive nature of the HEK293 cell line, we chose it as a platform for constructing glycan engineering based on the glycan elongation/branching pathway. Then, we analyze different glycosylation features based on the genetic characteristics of glycosyltransferases to further elongate and branch glycan structures.
    In this process, we offer RNA sequencing technology (RNA-seq) to predict the glycosylation capacity of cells. Gene KO and KI design techniques are also provided to add and remove glycan complexity branches.
  • Sublibrary strategy for glycan arrays of cells
    We develop a sub-library showing glycan characterization based on a set of isogenic cell groups. Among other things, we obtain specific glycans by eliminating similar types of glycoconjugates that display pathway extension/branching. In addition, we provide specific glycosyltransferases to enhance the elongation/branching features of the corresponding glycans.

Results: There are multiple types of glycosylation pathways in human cells and glycoproteins. One of these glycosylation pathways, based on glycan elongation/branching pathways, provides predictions of structural glycan features influenced by the presence or absence of individual genes. In addition, continuous analysis and refinement of glycosylation pathways increases insight into the non-redundant and competing functions of isozymes. To attain an in-depth analysis of glycosylation, the authors utilized CRISPR/Cas9 guide RNA libraries, KO targeting all human glycosyltransferase genes, and probed the stable site specificity of non-expressed genes.

  • Glyco-engineered cell constructs based on the glycan elongation/branching pathway can be used to analyze specific interactions between glycans and other biomolecules (e.g., between glycans and proteins).

  • Glyco-engineered cell constructs based on glycan elongation/branching pathways are useful for advancing the development of diagnostic tools based on glycan display array technology.
