High-throughput glycan screening based on liquid chromatography-mass spectrometry (LC-MS) is an effective method for glycan analysis. The system consists of an autosampler, liquid chromatography (LC), and mass spectrometry (MS) devices. The integrated system enables rapid and efficient separation and detection of a large number of glycan samples. Advanced automation software simplifies the entire process from sample injection to data analysis, minimizing human error and improving detection efficiency. LC separates glycans based on their physicochemical properties, while MS provides accurate quality metrics for identification, and the high sensitivity and selectivity of LC-MS accurately characterize complex glycan structures. This automated LC-MS-based solution enables researchers to explore a wide range of glycan modifications, patterns, and biological functions in a time- and labor-saving manner. It greatly accelerates glycan research and contributes to a better understanding of the role of glycans in various biological processes.
CD BioGlyco offers a high-throughput glycan screening service using an automated LC-MS-based solution. We employ state-of-the-art LC-MS instrumentation, coupled with automated sample handling and data analysis software, to achieve efficient and accurate glycan analysis. Our analytical process is as follows:
1) Pre-treatment of samples such as protein removal, removal of impurities, and desalination.
2) The sample is dissolved in a suitable solvent and injected into the LC system. Select a suitable column and set the composition and gradient conditions of the mobile phase to achieve a good separation of the glycans.
3) Introducing the separated glycans into the mass spectrometer detector. Depending on the type of MS used, the appropriate ion source and MS parameters, such as electrospray ionization (ESI), time of flight (TOF), etc., are set. The instrument is then run and the mass spectra are generated.
4) Acquisition and processing of MS data using relevant software.
5) The MS data obtained from the experiments are compared with a database of known glycans to preliminarily identify possible glycan structures and modifications. The structural features and modification sites of the glycans are further determined using software or manual extrapolation.
6) Confirmation of structural accuracy using other analytical techniques such as nuclear magnetic resonance spectroscopy (NMR).
