Results: In this work, researchers used differential cysteine alkylation and MS to determine the disulfide state of fibrinogen in human hepatocytes. Multiple analyses showed that fibrinogen is produced in multiple disulfide-bonded or covalent states and functions in multiple covalent forms. Further analyses revealed that disulfides are formed by the polymerization of fibrin and play a key role in the functioning of the protein. Notably, the polypeptide backbone of fibrinogen does not exist in a single covalent form in circulation but may be in hundreds of different covalent forms.
