Characterization of protein glycosylation has significance for understanding protein function and role in various disease states. The polarity of glycopeptides and oligosaccharides interact selectively with the polar stationary phase, which provides an opportunity for the application of HILIC in sample purification and analysis. HILIC has been successfully applied to several aspects of glycosylation analysis. PNGase F was used to perform glycan profile analysis of enzymatic glycan release, followed by derivatization with 2-aminobenzamide (2-AB) by HILIC-UHPLC, which is a powerful technique that can be used for the analysis of samples with well-known N-glycan profiles.
