Properties
Description
O-Glycosidase for core 1 (endo-α and Sialidase mix) is an enzyme mixture that contains endo-α-N-acetylgalactosaminidase and sialidase.
Molecular Weight
Endo-α (approx. 210 kDa), Sialidase (approx. 886652 kDa)
Unit Definition
One unit of the endo-α will produce 1 μmol of Galβ1-3GalNAc from Galβ1-3GalNAc α-pNP per minute at pH 5.0 at 37°C. One unit of the sialidase will produce produce 1 μmol of N-acetylneuraminic acid (Neu5Ac) from Neu5Ac-α-4MU per minute at pH 5.0 at 37°C.
Reaction Conditions
50 mM Sodium acetate, pH 5.0
Detection Method
O-Glycosidase for core 1 is a mixture of Endo-α and Sialidase. Where Endo-α and Sialidase are detected separately. The product at the end of the endo-α reaction is measured by UV spectrometer. The product released by sialidase is measured by HPLC.
Sensitivity
Heat sensitive
Concentration
Endo-α 0.1 U per vial (1 U/mL); Sialidase 1.0 U per vial (10 U/mL)
Form
Colorless to almost colorless clear liquid
Titer
Endo-α (≥0.1 unit/vial), Sialidase (≥1.0 unit/vial)
Buffer
20 mM Sodium phosphate, 150 mM NaCl buffer, pH 7.4
Applications
O-Glycosidase for core 1 (endo-α and sialidase mix) can be used for structural and functional analysis of glycan chains and to study the synthesis and modification of glycoproteins and glycolipids.
