- Physical and chemical properties
Isepamicin sulfate appears as a white or off-white powder. Isepamicin sulfate is easily soluble in water, however, it is not soluble in methanol and ethanol.
- Specific rotation
We measure the specific rotation of isepamicin sulfate using a spinometer. The specific rotation of isepamicin sulfate is usually between +100° and +120°.
- pH
The pH of isepamicin sulfate is tested using a pH meter. The pH of isepamicin sulfate is in the range of 5.5-7.5.
- Identification
- The sample to be tested is dissolved in water to which an anthrone solution is added. The color of the solution is used to identify the sample as isepamicin sulfate.
- We will dissolve the sample to be tested in water, to which barium chloride solution is added. When a white precipitate appears, hydrochloric or nitric acid is added. If the precipitate no longer dissolves, the sample is isepamicin sulfate.
- Content assay
High-performance liquid chromatography (HPLC): The aqueous solution of isepamicin sulfate is subjected to gradient elution using trifluoroacetic acid and methanol as mobile phases. Finally, the amount of isepamicin sulfate is obtained from the retention time of the main peak of isepamicin sulfate.
- Sulfate content
First, isepamicin sulfate is accurately weighed and dissolved in water. Then, the chromatogram of isepamicin sulfate is tested according to liquid chromatography. Finally, the amount of sulfate is calculated based on the area of the peaks in the chromatogram.
- Moisture content
A mixture of formamide-anhydrous methanol is used as a solvent to dissolve isepamicin sulfate, which is then assayed for moisture content using volumetric titration.
- Cauterization of residue
After weighing isepamicin sulfate, we place it in a crucible. Then, it is heated until it is completely charred. Finally, it is transferred to a desiccator and then it is weighed to get the residue weight of isepamicin sulfate.
- Abnormal toxicity test
First, isepamicin sulfate is dissolved in sodium chloride injection, and then the resulting solution is injected intravenously into mice. If the mice die within a specific time, isepamicin sulfate is abnormally toxic.
- Antihypertensive substance
We inject a sodium chloride solution of isepamicin sulfate into an animal and the blood pressure of the animal before and after the injection will be recorded. The changes in blood pressure are analyzed to determine whether isepamicin sulfate contains antihypertensive substances.

Results: The authors used reversed-phase HPLC to analyze isepamicin sulfate. Wherein, the authors used a set of tandem columns to separate and purify isepamicin sulfate at 10°C. Then, the impurity analysis of isepamicin sulfate was also evaluated using different standard curves. For example, a standard curve of 1.6 mg/ml can be used to analyze the amount of isepamicin sulfate. Whereas, a standard curve of 8.0 mg/ml can be used to detect impurities of isepamicin sulfate.
