Sialic acid glycoengineering is increasingly applied in the field of glycoscience and beyond. Chemically modified analogs of N- N-acetylmannosamine (ManNAc), the biological precursor of sialic acid, or sialic acid analogs are introduced into the metabolic sialic acid biosynthesis pathway resulting in their incorporation into sialoglycans.
Over the last two decades, more than 20 unnatural mannosamines have been introduced as suitable substances for metabolic oligosaccharide engineering. These ManNAc analogs can be divided into two groups: aliphatic and bioorthogonal analogs. In aliphatic analogs, the N-acyl function is elongated by one or more methylene groups. Metabolic oligosaccharide engineering with aliphatic ManNAc analogs introduces novel biological characteristics to cells and consequently influences their behavior in multiple ways. Bioorthogonal analogs carry modifications of the N-acyl side chain that are both absent in living systems and utilizable for further chemical reactions. These reactive groups are applied, for example, to label sialylated glycans with a fluorescent dye.
