Role of Mannosamine

Mannosamine is an amino derivative of mannose. It can serve as an inhibitor to prevent the incorporation of glycosylphosphatidylinositol (GPI) into proteins. Mechanistically, it reduces the expression of GPI-anchored proteins and converts the proteins into unpolarized secretory products by consuming GPI precursor lipids with strong transport capacity. In addition, mannosamine also inhibits aggrecanase-mediated cleavage of aggrecan and relevant changes in the physical properties and biochemical composition of articular cartilage, which is an important part of the pathophysiology of osteoarthritis.

For amino-saccharides, the traditional detection methods include GC, HPLC, and IR. However, in GC, the sample pretreatment process is complicated and derivatization is required. The sensitivity of IR is so low that the detection effect is difficult to reach the expectation.
