Mannosamine is an amino derivative of mannose. It can serve as an inhibitor to prevent the incorporation of glycosylphosphatidylinositol (GPI) into proteins. Mechanistically, it reduces the expression of GPI-anchored proteins and converts the proteins into unpolarized secretory products by consuming GPI precursor lipids with strong transport capacity. In addition, mannosamine also inhibits aggrecanase-mediated cleavage of aggrecan and relevant changes in the physical properties and biochemical composition of articular cartilage, which is an important part of the pathophysiology of osteoarthritis.
