Melibiose is an analog of Trehalose, which is linked by α-1,6-glycosidic bonds to a molecule of glucose and a molecule of galactose. HPLC is the most commonly used technique for the determination of sugar content in samples. The method shows the advantages of precision, selectivity, and sensitivity. Our detectors include refractive index detection (RID), ultraviolet (UV), and evaporative light scattering detector (ELSD). Meanwhile, melibiose labeling is suitable for detection by UV detector, which improves the sensitivity and accuracy of detection. After HPLC analysis, the samples are analyzed in tandem with ESI-MS for sample characterization, and tandem mass spectrometry is used to attribute and determine the structure of the sugar chain from the fragmentation feature profiles derived from multistage cleavage of the sugar chain.

Results: In the study, myoglobin (MB) glycosylated with mel is analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE), showing a mixture of products with different molecular masses and degrees of cross-linking. The mel under dry conditions (MWG) condition favored the generation of products with a higher degree of cross-linking, a richer mel loading, and different antigenic properties.
FTIR analysis showed that the protein structure is significantly altered after glycosylation under both conditions. The differences in physicochemical properties between MWG and conventional glycation condit glycosylation products are confirmed by absorption and fluorescence properties.
Analysis of the spatial distribution within the proteins of known advanced glycation end-products (AGEs) using LC-MS/MS also revealed differences in the antigenic properties of melibiose-derived advanced glycation end-product (MAGE), showing specific MAGE-modified blood proteins. Further studies of the spatial modifications of immunoglobulins and serum albumin are highly correlated with MAGE-modification-associated cancers and diabetic diseases.
