The pH value of metronidazole and glucose injection should be 4.5~6.0.
- Determination of nitrite content
Nitrite content is determined by ion chromatography (5 mmol/L potassium hydroxide solution as eluent). Sodium nitrite is used as the control, and the content of nitrite in the test material is calculated by peak area. The content of nitrite should not exceed 0.8% of the labeled amount of metronidazole.
- Detection of relevant substances
HPLC with light protection is used to detect a wide range of substances.
- 5-Hydroxymethylfurfural assay
We take the appropriate amount of 5-hydroxymethylfurfural control product and configure it into the control solution. After that, we inject the test and control solution into the liquid chromatograph, record the chromatogram, and calculate the content of 5-hydroxymethylfurfural according to the peak area.
- Osmotic pressure molar concentration
The osmotic pressure molar concentration of the test product is checked. The osmotic pressure molar concentration ratio should be 0.9~1.1.
- Heavy metals
We take an appropriate amount of the test material and place it in a water bath to evaporate. After cooling, we add acetate buffer and water to a certain amount and test the heavy metal content.
- Others: Harmful element residue test, endotoxin test, insoluble particle test, sterility test, etc.
- Determination of content
- Glucose: We measure the optical rotation of the test material, and the content of glucose is calculated.
- Metronidazole: We measure the content of metronidazole by HPLC using methanol-water as the mobile phase.

Results: In this study, an LC-MS/MS method was developed for the determination of metronidazole, chloramphenicol, and 10 sulfonamides. The method is sensitive, and rapid, and can be successfully applied to the simultaneous determination of multiple substances. This study provides an effective method for the analysis of metronidazole and glucose injection.
