One of the most important functions of an oligosaccharide library is to develop oligosaccharide microarrays, so its construction is very important. Results: The authors describe a synthetic method for the construction of heparan sulfates (HS) oligosaccharide libraries. The structure is based on the use of modular disaccharides to form selectively protected tetrasaccharides. These tetrasaccharides have been subjected to selective chemical modification such as regioselective O- and N-sulfation and selective desulfation and enzymatic modifications. A total of 12 different HS derivatives were obtained. A library of 47 HS oligosaccharides has been developed and the resulting compounds have been synthesized to create HS oligosaccharide microarray, using tetrasaccharide backbone compositions with various compositions. The ligand requirements of many HS-binding proteins, such as fibroblast growth factor and chemokines, were examined using this array.
