The method utilizes chemical derivatization techniques to increase the sensitivity for the determination of GM1 in samples. The method uses reversed-phase ultra-performance liquid chromatography (UPLC) for chromatographic separation and tandem mass spectrometry in multiple reaction monitoring (MRM) modes to enhance detection, sensitivity, and specificity. Taking plasma samples as an example, our analysis process is as follows.
Preparing working plasma samples using plasma and internal standard working solution. After cooling and vortexing, the mixture is centrifuged and the supernatant is transferred to an autosampler vial with a micro-insert for LC/MS analysis.
After chemical derivatization, the sample is injected into a UPLC column. Elution and analysis are carried out according to a certain analysis procedure. The introduction of an electrospray ionization source is required during the analysis because all derivatized monosialoganglioside components are eluted and detected sequentially.
Ganglioside quantification is performed on plasma samples using a calibration curve based on seven levels. For each of the three monosialogangioside species, the total peak area is calculated by summing the IS-normalized peak areas of the individual components based on a single injection.
