The structurally simple GM3 contains a terminal sialic acid. GM3 regulates the function of several receptors involved in angiogenesis. GM3 also reduces proliferation and enhances apoptosis of rapidly proliferating neural stem cells. In addition, GM3 has the effect of inhibiting angiogenesis inducers. We offer ultra-high performance liquid chromatography combined with electrospray ionization triple quadrupole tandem mass spectrometry (UPLC-ESI-MS/MS) for the determination of GM3 in infant formula milk powder and whey protein concentrate. Stock solutions of graded concentrations are prepared by dissolving GM3 in methanol. GM1 is diluted with 70% methanol to a certain concentration as an internal standard (IS).
Sample pretreatment is carried out in two steps, including sample extraction and clean-up by solid-phase extraction.
The UPLC system consists of an ultra-high-performance liquid chromatograph equipped with an autosampler. The chromatographic separation of GM3 is performed on a chromatographic column. Separation is carried out strictly according to the elution procedure. Measurements are performed in ESI mode on a triple quadrupole mass spectrometer equipped with an electrospray ionization (ESI) source.
Standard calibration curves for ganglioside molecules are established according to the IS method. The peak areas of the spot ion chromatograms of the GM3 molecule are calculated for calibration levels within the calibration range. All experiments are performed in triplicate. The concentration of GM3 molecules in the sample extracts is obtained based on the linear regression equation. This concentration is further multiplied by its corresponding percentage to obtain the true value.
