Overview of Monosialoganglioside 4 (GM4)

GM4 is a monosialoganglioside found mainly in the central nervous system and was found to be a major component of myelin gangliosides. It has also been identified as a specific marker for human myelin and oligodendrocyte nuclei. In the chicken cerebellum, GM4 is associated with astrocytes but not myelin. The appearance of GM4 in extraneural tissues was first reported in egg yolk and chicken thymus. This ganglioside has also been detected in mouse erythrocytes, rat kidneys, chicken embryo liver, chicken liver, and frog liver. Among these tissues, GM4 was found to be the major ganglioside in egg yolk, chicken embryo liver, and frog liver.

Secondary ion mass spectrometry (SIMS) spectra of gangliosides are recorded on a quadrupole mass spectrometer in negative and positive ion modes. The sample dissolved in chloroform-methanol is loaded onto the SIMS target tip, analyze by the program and a scan is acquired to obtain a spectrum.

  • NMR

For NMR analysis, ganglioside is exchanged with D2O, lyophilize, exchange again with CD3OD, and evaporate to dryness. The 1H-13C correlation of the heteronuclear single bond is detected, and the 1H heteronuclear multiple bond linkage is detected.
