CD BioGlyco has extensive experience with NGL display arrays based on the Glycan Display Platform. Immobilization of NGL on solid surfaces or incorporation into liposomal formulations is an important step in the construction of NGL Display Arrays. For this purpose, we have studied the effect of various methods on NGL immobilization. Based on our research experience, we offer high-quality NGL immobilization services. Our goal is to immobilize NGL in liposomal formulation or on solid supports in a controlled and stable manner.
We chemically attach the NGL to the solid support through covalent bonding. Various functional groups on the solid support and NGL are used for the affixation such as amino group and carboxyl group.
We also employ non-covalent interactions to immobilize the NGL to the solid support. Common methods include hydrophobic interactions, electrostatic interactions, or specific binding interactions. For example, a lipid anchor with a hydrophobic portion is immobilized by adsorption onto hydrophobic solid support via hydrophobic interactions.
NGL is used in inhibition assays when doped into liposomes in a multivalent state. We immobilize NGL in a multivalent manner in which multiple copies of NGL are displayed on a solid support. In this way, the affinity and binding of NGL to specific proteins are enhanced. This method also mimics the multivalency present in natural carbohydrate recognition events.
The choice of the NGL immobilization method depends on the structure of the NGL, the solid support material, the desired stability, and the application. During the immobilization process, we optimize and examine the effects of NGL concentration and structure, solid support material, connecting molecules, durability, and other factors. Stable and reliable NGL immobilization is ultimately achieved through continuous optimization.

Results: This paper describes a method for preparing a microarray of lipid-linked oligosaccharide probes by covalent immobilization. The microarray combines NGLs and oligosaccharide arrays with each other. The microarray was employed to examine the oligosaccharide binding properties of the C-type lectin-like protein dectin-1, and the results showed that dectin-1 binds to 10 aggregates or longer β1,3-linked oligoglucose sugars. This method provides an idea for NGL immobilization and NGL-based analysis.
