Phaseolus vulgaris Lectin (PHA) Production Service at CD BioGlyco

- PHA extraction and purification
- First, we crush and homogenize the Phaseolus vulgaris. Immediately after that saline or buffer is added to it as an extractant to dissolve it. Then the supernatant is collected by ultrasonication and centrifugation operation. Finally, the crude product of PHA is obtained by freezing and drying.
- We purify the crude product further by graded precipitation using ammonium sulfate and Ion Exchange Chromatography techniques. Using sodium dodecyl sulfate-polyacrylamide gel electrophoresis and mass spectrometry to characterize the protein and determine its purity.
- PHA property assay
- Hemagglutination activity
The hemagglutination method is utilized to detect the agglutination activity. Buffer is added to the wells of a hemagglutination plate, and different amounts of lectin samples are added to the wells separately, followed by erythrocyte suspension. The coagulant activity of the lectin is observed under the naked eye or microscope.
- Thermal stability
PHA is dissolved in buffer heat treatment is carried out in a water bath and the hemagglutination activity of the lectin at different temperatures is detected using the hemagglutination method.
- Metal ion binding
PHA is dissolved in buffer and centrifugation. Different metal ion solutions are added separately and the hemagglutination activity is detected using the hemagglutination method.
- Sugar binding specificity
We add separate solutions of different sugars to the PHA solution and assay the hemagglutination activity using the hemagglutination assay.

- Results: Zihua snap bean was characterized via sodium dodecyl sulfate-polyacrylamide gel electrophoresis, liquid chromatography, mass spectrometry, infrared spectroscopy, and fluorescence spectroscopy for its physicochemical properties and biological activities. The results showed that (I) glucose, N-acetyl-d-glucosamine, galactose, N-acetyl-galactosamine, fructose, sucrose, maltose, alginate, and lactose did not bind well to the lectin. (II) The hemagglutinating activity of the lectin showed a strong dependence on Mn2+ and Ca2+. (III) Purple-flowering bean curd agglutinin had good thermal and pH stability. (IV) The untreated lectin was relatively stable to pepsin and trypsin activity, but heat treatment significantly reduced digestive stability in vitro. In addition, the lectin was inhibitory to the tested bacteria (S. aureus, E. coli, B. subtilis) and also had an inhibitory effect on bacterial growth.

Applications
- PHA is highly thermostable, and it is used to develop an efficient yet safe plant-derived biopesticide.
