• Extraction and purification services

In seeds and tubers of the plant, PTL is usually present. First, we are going to crush the seed or root of P. tetragonolobus and make a homogenous pulp. To extract it, add a solution of inorganic salts to it. The mixture is processed by low-temperature, high-speed centrifugation, and then we collect the supernatant and freeze-dry it to obtain the crude product of PTL.

To purify crude PTL, we use Gel-filtration Chromatography and Ion Exchange Chromatography to obtain high-purity PTL.

• Property testing services
  • Agglutination activity test
    We test the hemagglutination activity of PTL using the hemagglutination method or tube method. In the hemagglutination method, a sample of the lectin and a suspension of red blood cells are added to the wells of a hemagglutination plate, which is left to stand, and the hemagglutination effect of the lectin is observed under a microscope. We also offer red blood cell modification services to enhance the sensitivity of red blood cells to the lectin.
  • Protein content assay
    With animal serum as the standard solution, we use a spectrophotometer to measure the absorbance of the lectin solution and calculate the protein content.
  • Sugar binding assay
    After labeling different sugar complexes with radioisotopes, they are incubated with PTL. The conjugates are separated with the help of methods such as centrifugation or filtration. The amount of labeled glycoconjugates as well as the binding constants are calculated by detecting the intensity of the radiolabel in the precipitate.
  • Stability assay
    Thermal stability: Using the hemagglutination method, test the hemagglutination activity of PTL at different temperatures to get the result of thermal stability.
    Acid-base stability: Using different pH buffers to dissolve the PTL, test the agglutination activity of the lectin in different solutions to get the results of its acid-base stability.
