Description
Recombinant keratanase II from Bacillus circulans cleaves the sulfated N-acetylglucosaminidic β1-3 linkage to galactose included in keratan sulfate.
Unit Definition
One unit will produce 1 umol of reducing oligosaccharides from shark cartilage keratan sulfate polysaccharide per minute at pH 6.0 at 37°C.

A 50 ul mixture of recombinant keratanase II and shark cartilage keratan sulfate is incubated at 37°C for 15 min, and the released oligosaccharide reducing end is determined by a modified Somogyi-Nelson method.
