The yeast expression system has the advantages of easy genetic manipulation and rapid growth in prokaryotes, combined with the molecular genetic characteristics of higher eukaryotes. It is well suited for expressing lectins. Yeast potentially generates soluble, correctly folded recombinant proteins. CD BioGlyco mainly provides two classes of yeast expression systems for the Expression and Production of Recombinant Lectins: the Pichia pastoris expression system and the Schizosaccharomyces pombe expression system. Meanwhile, we offer other yeast expression systems such as Arxula adeninivorans, Hansenula polymorpha, Kluyveromyces lactis, and Yarrowia lipolytica. We perform bulk preparation of recombinant plasmids by cloning the PCR amplification products into suitable vectors for transformation—selection of suitable yeast hosts for rolling circle replication (RCR) characterization for multicopy subclones. Select multiple plants with multiple copies for a small amount of induced expression, our technician will send the expression supernatant sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) to your email address. According to your needs, select the best expression situation to amplify the expression. The supernatant of fermentation broth is processed and purified, and the purified product is provided with various indexes for testing.

Results: The electrophoretic profiles of both n-ArtinM and y-ArtinM are of a single 16 kDa band. The molecular mass is similar and the tetramer is organized by examining the molecular mass and structure. The carbohydrate binding sites of both lectins were analyzed by glycan microarray, both with primary and secondary subsites that interacted with complex glycans. Notably, y-ArtinM migrated more efficiently than n-ArtinM in vitro assays. Meanwhile, y-ArtinM also stimulated neutrophils to release myeloperoxidase (MPO), which was induced at a level similar to that of n-ArtinM. In summary, y-ArtinM was similar to n-ArtinM in terms of subunit arrangement, oligomerization, carbohydrate binding specificity, and immunological properties.
