Rhamnan is a polysaccharide that is polymerized from monosaccharides such as rhamnose. Since pure rhamnose crystallizes in two forms, α-type and β-type, the backbone structure of rhamnan also shows these two forms. Rhamnan has a variety of biological activities, including antioxidant, antibacterial, antiviral, antitumor, hypolipidemic, and immune regulation. Studies have shown that rhamnan can promote cell regeneration and repair, and has a protective effect on many organs, including the digestive tract, cardiovascular system, and nervous system. In addition, it can also be used as an additive in cosmetics and food. Therefore, rhamnan has a broad application prospect in medical and biological fields.

Results: Ultraviolet-visible spectrum (UV-Vis) spectra of exopolysaccharides (EPS) solutions were recorded on a UV spectrophotometer scanning from 200 to 800 nm, aiming to estimate the purity of the purified components. Infrared spectroscopy (IR) spectra of the purified EPS were recorded on a Nicolet 380 infrared spectrometer at the wave number range of 4000-400 cm−1 using a KBr pellet method. 1H NMR spectra of EPS were recorded on a Bruker Avance 600 spectrometer, and 13C NMR spectra of EPS were recorded on a Bruker Avance 400 spectrometer.
Almost all of the IR spectra, 1H NMR, and 13C NMR spectra can be used for the analysis of the glycosidic bond configuration of polysaccharides (Fig.2). From the plot, it is clear that α-glycosidic linkage is the main linkage between the monosaccharide units in EPS has both α- and β-glycosidic linkages.
