Properties
Description
Sialidase agarose from Arthrobacter ureafaciens is an enzyme-immobilized agarose that cleaves the glycosidic linkages of neuraminic acids.
Unit Definition
One unit will produce 1 μmol of N-acetylneuraminic acid (Neu5Ac) from Neu5Ac-α-4MU per minute at pH 5.0 at 37°C.
Source
Arthrobacter ureafaciens
Substrate (Enzyme)
Micrococcus luteus
Reaction Conditions
50 mM Sodium acetate, pH 5.0
Detection Method
The mixture of sialidase-immobilized agarose and Neu5Ac-α-4MU in 200 μL is incubated at 37°C for 10 min. Adding a 4-fold volume of 0.1 M sodium carbonate buffer, pH 10.0 to quench after the enzyme reaction. The released 4-MU is determined by HPLC.
Sensitivity
Heat sensitive
Concentration
0.1 mL gel/0.2 mL (1:1, v/v slurry)
Form
White to gray cloudy liquid
Buffer
20 mM Sodium phosphate buffer, pH 7.4
Preservative
0.02% [w/v] Sodium azide
Applications
Sialidase agarose from Arthrobacter ureafaciens not only immobilizes the enzyme neuraminidase on its surface, but also captures and purifies proteins or viruses bound to neuraminic acid through affinity interactions.
