LSTa, LSTb, and LSTc are the dominant constituent of complex human milk oligosaccharides( HMOs). They effectively inhibit the adhesion of bacteria to epithelial surfaces, thereby blocking the first stage of the infection process, and are considered to play a major role in immune defense against viral infections and bacteria in the gastrointestinal tract of infants.

Some animal milk oligosaccharides are used as functional foods, such as replacing human milk as the bioactive ingredient in infant milk powder. Donkey milk has been considered to have this potential, studies have shown that donkey milk contains sialylated milk oligosaccharides such as LSTa, LSTb, and LSTc.
Many oligosaccharides are synthesized by classical chemical methods or only enzymatic methods. For the oligosaccharide structure with a clear regional and stereochemical layout and specific substitution mode, it is necessary to improve the efficiency of each step and try to simplify the purification step to improve the yield. In addition, using these two procedures to synthesize in synergetic mode is also a good method to obtain sialylated structures. One of the most suitable general methods for this situation is block synthesis. In recent years, some combination methods of chemical and chemoenzymatic synthesis for LST derivatives are reported.
