Background

A large number of proteins in eukaryotes are glycosylated and they play a variety of roles in physiological functions like molecular recognition. Therefore, glycosylation analysis is one of the indispensable approaches for developing antibody drugs and discovering biomarkers. Nowadays, various detection methods are emerging, among which LC-MS can detect and clarify the low abundance metabolites in the research of natural products. However, mass spectrometry (MS) can not provide information about the spatial and temporal distribution of metabolites in biological samples.

Imaging mass spectrometry (IMS) complements the traditional metabolomics and chemical analysis by combining qualitative and quantitative molecular information with spatial and temporal information. Moreover, MALDI-imaging mass spectrometry (MALDI-IMS) is a powerful technology, which can be used to study the spatial distribution of thousands of molecules in the whole tissue section through an experiment. MALDI-IMS has been used in many substances research, including metabolites, drugs, lipids, peptides, and proteins. However, its application in glycans is rarely reported.
