Steviol glycoside is a kind of glycoside extracted from the leaves of Stevia rebaudiana, the main component is stevioside (C38H60O18), usually accompanied by rebaudioside A, B, C, D, F, dulcoside A, sweet tea glycoside and stevia disaccharide glycoside and other glycoside components. Steviol glycoside is often used as a new natural sweetener, and also steviol glycoside is a pharmaceutical excipient with anti-diabetic, antihypertensive, anti-inflammatory, antibacterial, and other functions.

  • Identification
    Thin layer chromatography: The sample and the standard of steviol glycoside are dissolved in anhydrous ethanol, and then spotted on the same silica gel thin layer plate and unfolded. By comparing the spot positions of the sample and the standard, it can be identified whether the sample is steviol glycoside or not.
  • Properties
    Steviol glycoside is a white or off-white crystal or powder, soluble in a mixture of ethanol and water.
  • Specific rotation
    The specific rotation of steviol glycoside is determined after it has been fully dissolved in a mixture of ethanol and water and should be between -30° and -40°.
  • Acidity
    The acidity of steviol glycoside can be measured using a pH meter. The pH of steviol glycoside should be 4.5 to 7.0.
  • Content assay
    Titration: We titrate a sample solution of steviol glycoside using phenolphthalein as an indicator solution and an alcoholic solution of potassium hydroxide as a titrant. The titration is stopped when the solution turns red. The content of steviol glycoside (C38H60O18) in the sample can be calculated from the amount of titrant used.
  • Solvents (methanol and ethanol)
    Liquid chromatography: We measure methanol and ethanol in steviol glycoside by liquid chromatography method which uses polyethylene glycol as a stationary solution and butanol as an internal standard solution. The amount of methanol and ethanol can be calculated from the internal standard method by peak area.
  • Loss on drying
    At higher temperatures, we dry steviol glycoside. After complete drying, the weight loss of steviol glycoside should not exceed 5.0% of the weight of the dried product of steviol glycoside.
  • Scorch residue
    The amount of residue remaining after scorching of steviol glycoside is not to exceed 0.1 percent.
  • Heavy metals
    The residue from the burning of steviol glycoside is treated with acid, to which acetate buffer and thioacetamide test solution are added. If the steviol glycoside contains heavy metal impurities, the whole solution becomes darker in color.
  • Lead
    The amount of lead contained in steviol glycoside can be calculated from the absorbance at 283.3 nm, which is determined by atomic absorption spectrophotometry, provided that steviol glycoside is completely digested.

Results: Known as a low-calorie sugar substitute, steviol glycoside is a natural sweetener used in foods and beverages around the world. In this paper, the authors developed a more excellent method to analyze the content of steviol glycoside. Using HPLC in combination with a pretreatment method, the authors have measured the steviol glycoside equivalents in dairy products directly. During the experiment, the addition of MeOH and NaH allowed for a more reliable determination of the total amount of steviol glycoside in the samples. Moreover, the pre-treatment method used by the authors allowed the assessment of steviol glycoside levels with high accuracy, linearity, and precision.
