Fat is an indispensable nutrient and basic metabolite for our life, and it is the main form of energy storage in the body. Fat is composed of a complex mixture of FAs, including saturated, monounsaturated, and polyunsaturated compounds with various carbon chain lengths. Since the role of FAs in the body depends on the structure, it is necessary to conduct a detailed analysis of the composition of FAs.
FAs are the main components of lipids, and the physical, chemical, and physiological properties of lipids mainly depend on the composition of FAs. Generally, the composition of FAs is analyzed by gas chromatography (GC). Therefore, it is necessary to convert TFAs into derivatives with less polarity and higher volatility. After lipid extraction, TFAs are usually converted to their methyl ester (FAs methyl ester, FAME) before chromatographic analysis. Boron trifluoride methanol solution is the commonly used derivatization reagent for FAs methyl esterification. One of the key issues of analyzing FAME from TFAs is to choose a suitable chromatographic column to separate them, which mainly depends on the objective of the analysis, and the internal standard method is usually used for quantification.
