Tumor-associated glycan antigens are polysaccharides that are widely expressed on the surface of a variety of tumor cells, of which the globo series of antigens are representative of tumor-associated glycan antigens, which are widely expressed in a variety of malignant tumor cells, including breast cancer. Gb5 antigen in the globo series is a glycoprotein related to the glomerular basement membrane, which is one of the important components of the glomerular basement membrane and participates in the maintenance of glomerular filtration function. The structure of the Gb5 antigen consists of a transmembrane region, an extracellular region, and an intracellular region. Gb5 antigen is involved in the regulation of glomerular epithelial cell proliferation, differentiation, and polarity, and is widely expressed in tumor cells. In addition, Gb5 antigen has been applied in the Development of Tumor-Related Vaccines.

CD BioGlyco has established a Vaccine Development Service Platform and provides vaccine development, antigen production, and other related services. Among tumor-related antigens, we have mastered the Production Process of Globo Antigens. Among them, Gb5 is synthesized as follows:
This method we offer employs three glycosyltransferases: galactosyltransferase, acetylgalactosaminyltransferase; and fucosyltransferase. The enzymatic reaction of these enzymes is utilized to insert glycosidic bonds to synthesize the Gb5 antigen.
Modification of the ceramide backbone using the modular assembly of the substrates β-lactosylsphingosine and sphingolipids is an effective method for synthesizing Gb5 antigen.
We use glycosyl phosphates as donors for the automated synthesis of Gb5 antigen on solid carriers in a strictly linear strategy and purification by column chromatographic separation.
In our vaccine development services, we use the Gb5 antigen as the core substance in combination with different surfactants, adjuvants, stabilizers, and other active substances for vaccine development. We offer in vivo and in vitro assays to test the effectiveness of the vaccine.
