Vicia villosa Lectin (VVL) Production Services at CD BioGlyco

- VVL extraction and purification
First, we crush the Vicia villosa seeds. Secondly, the extraction solution is used for extraction. Finally, after centrifugation, the supernatant is collected to obtain the crude VVL.
We use Affinity Chromatography to purify VVL. The monosaccharide is used as the affinity adsorbent, and after attaching the lectin crude to the affinity chromatography column, the lectin is eluted step by step using a salt solution and collecting the eluent, dialyzing, and lyophilizing to obtain the pure product of VVL.

- VVL property assay
- Purity analysis
VVL is stained and analyzed for purity using polyacrylamide gel electrophoresis.
- Agglutination activity analysis
Using the hemagglutination test, VVL is mixed with erythrocyte suspensions from different animals, left to stand, and the coagulation effect of the lectin in each mixture is observed under a microscope.
- Protein content analysis
Utilizing a spectrophotometer to detect the absorbance of VVL under the standard vertebrate solution, the protein content is calculated from the absorbance curve.
- Sugar binding property analysis
Using different sugar solutions to mix well with VVL, the binding capacity between VVL and sugar is detected according to the radioisotope labeling method.
- Metal ion binding assay
Dissolve the lectin with buffer and after dialysis and centrifugation process, add different metal ion solutions to it and assay the hemagglutination activity using hemagglutination assay.
- Stability assay
Detection of hemagglutination activity of VVL at different temperatures and pH values using hemagglutination assay to get its stability results.
