Vigna radiata Lectin (VRA) Production Services at CD BioGlyco

Lectins are glycoproteins that are capable of agglutinating human and animal red blood cells. Natural lectins are mainly derived from plants, animals, and fungi.   • Separation extraction and purification

Firstly, we crush Vigna radiata and extract VRA by using salt solution as the extracting solution. Secondly, we add a certain amount of phosphate buffer into it to improve the extraction efficiency of the lectin. Finally, the crude extract of VRA is obtained by centrifugation, sedimentation, and filtration.

The crude extract of VRA is double-salted with ammonium sulfate to collect the precipitate. After extraction and dextran gel filtration, high-purity VRA is obtained.

  • Property analysis
    • Structural detection
      To detect different levels of structure of VRA using infrared spectroscopy and circular dichroism spectroscopy.
    • Agglutination activity assay
      The agglutination activity of VRA is assayed by coagulation assay. Blood erythrocyte suspensions of different animals are used andVRA is added to them separately. After standing, the coagulant activity is observed.
    • Molecular weight assay
      Sodium dodecyl sulfate-polyacrylamide gel electrophoresis is an effective method for detecting its molecular weight and shows the protein bands clearly.
    • Optimal extraction buffer solution assay
      Different buffers with different concentrations and pH are used for extraction under the same conditions, and the agglutination activity of the extracts is determined separately to determine the optimal extraction buffer solution.
    • Stability test
      The agglutination activity is measured at different temperatures or different pH values to determine the thermal and acid-base stability.
    • Sugar binding assay
      Dissolve VRA in buffer add different types of sugar to the solution separately and assay the hemagglutination activity using the hemagglutination method.
