Sialic acids are alpha-keto acid sugars located at the end of the glycan structure of glycoproteins and glycolipids. They generally exist on the periphery of oligosaccharides via α2,3-, α2,6-, α2,8- and α2,9-glycosidic bonds, and can also be attached to glycoproteins in the form of linear homopolymers. Due to its unique terminal position and electronegativity, sialic acids control cell activation, differentiation, transformation, and migration. In tumor biology, sialic acid promotes immune escape, tumor proliferation and metastasis, tumor angiogenesis, resistance to apoptosis, and cancer therapy. Therefore, developing strategies of sialic acid analysis in cells and tissues can help to better understand the relevant physiological and pathological processes.
Currently, several bio-affinity-based methods have been developed to determine specific sialic acids, sialic acid glycans, and their biomodifications on cells and tissues, including sialic acid-specific lectins, anti-sialic acid-specific antibodies, and recombinant sialic acid-binding proteins. Lectins are sugar-binding proteins mainly used to analyze the expression of specific sialic acids on the cell surface. For example, Sambucus nigra lectin (SNA) recognizes α2,6-linked sialic acid residues. Antibodies are used to study cell-surface sialic acids and sialic acid glycans. Recombinant sialic acid-binding proteins measure specific sialic acids, sialic acid glycans, and their biological modifications on cells and tissues.

We analyze sialic acid on the cell surface using anti-N-acetylneuraminic acids (Neu5Ac) antibodies, anti-N-glycolylneuraminic acids (Neu5Gc) antibodies, anti-O-acetylated sialic acids antibodies, anti-polysialic acid antibodies, anti-gangliosides antibodies, anti-sialyl Lewis a (SLea) antibodies, anti-sialyl Lewis x (SLex) antibodies. We use sialic acid-specific lectins to analyze cell surface-specific sialic acids, sialyl glycans, and their biomodifications by histochemistry, flow cytometry and fluorescence microscopy, and lectin microarrays. In addition, we also provide sialic acid analysis services using recombinant sialic acid-binding proteins, such as sialic acid-specific recombinant microproteins, recombinant Siglecs-fragment crystallizable (Fc) fusion proteins, and recombinant endoenuraminidase-green fluorescent protein (GFP) fusion proteins.
