  • Production of sialic acid-binding lectin

We use specific bacteria as hosts to construct lectin expression vectors and genetically engineer them to produce sialic acid-binding lectin in their C-terminal structural domains using galactose-binding lectin as a precursor. In this process, Gly and Ser residues act as flexible linkers for constructing tandemly repetitive sequences of sialic acid-binding lectin plasmids.

  • Purification of sialic acid-binding lectin

We purify sialic acid-binding lectin using Affinity Chromatography. After purification, the lectin is purified again by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and lactose-agarose column. Finally, high-purity sialic acid-binding lectin is obtained.

  • Analysis of sialic acid-binding lectin
    • Coagulant activity analysis
      Use buffer to dissolve the lectin, add suspensions of different bacteria or animal erythrocytes to it, and observe the agglutination activity of the lectin on different bacteria and animals under the microscope.
    • Sugar binding assay
      Different sugar solutions are added to the lectin samples, and each mixture is analyzed for affinity between the lectin and the sugar, and the inhibitory activity of the sugar on the lectin, using a lectin immobilization column.
    • Glycoprotein binding assay
      We utilize glycoprotein imprinted microarrays to detect the binding signals between the lectin and different glycoproteins by a sudden field fluorescence scanner, which is used to test the binding ability of the lectin to different glycoproteins.
