We synthesize α2-3-linked sialic acid-binding lectin by constructing expression vectors in specific cells using genetic engineering methods.
In this case, salivary acid residues are present at the outermost ends of the sphingolipids, and N- and O- glycans bind directly to the salivary acid residues to obtain glycans with terminal α2-3-linked salivary acid in N- and O-linked. On the other hand, in the above process, the doping-protection-deprotection method, which occupies the N-terminus of the glycan, prepares a glycan with terminal α2-3-linked sialic acid (Neu5Ac)-in O-linked.
- α2-3-linked sialic acid-binding lectins purification
We utilize a Gel Filtration column and an Affinity Chromatography column to purify the lectin. Pure lectin is obtained by collecting the product with a strong absorption peak in a split tube, as well as using a concentration and drying operation.

- Agglutination activity
The hemagglutination method is used to test the agglutination activity of this lectin. The process is as follows: add the red blood cell suspension of the animal in the lectin solution, and after a while, observe the coagulation state in the mixture under the microscope or directly with the naked eye to get the agglutination activity of the lectin.
- Molecular weight and structure
The subunit composition, protein content, and molecular weight of the lectins are analyzed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis and ultraviolet spectroscopy.
- Sugar binding properties
The sugar-binding capacity of the lectin is analyzed using glycoconfiguration microarrays and affinity chromatography, and the binding capacity of different sugars are also compared.
