• Extraction and purification of AIA
  • Extraction
    We suspend the ground pineapple honey seeds in a NaCl solution. Next, we stir the solution, filter it, and centrifuge it. Rivano is added to the supernatant, the solution is again stirred and filtered, and centrifuged to remove the precipitate. Then KBr is added to remove excess Rivano, the residue is filtered and removed, and the supernatant is lyophilized to obtain the crude product of AIA.
  • Purification
    We use a Gel Filtration Column and NaCl solution to purify AIA and will pass the ultraviolet and coagulant activity test after the active fraction.
• Determination of the properties of AIA
  • Homogeneity: We determine the homogeneity of lectin by the disc gel electrophoresis method.
  • Molecular weight: The molecular weight of the purified lectin is determined by the gel column.
  • Carbohydrate content: To identify the amount of total neutral glycans and individual glycans, we analyze the lectin samples with HCl in hydrolysis and analyze them by gas-liquid chromatography.
  • Metal analysis: The purified lectin is dissolved in brine and we determine the metal content using an atomic absorption spectrophotometer hollow cathode lamp.
  • pH effect: The lectins are placed in the presence of different buffers to test their activity.
