Biomedical Applications of Carboxymethyl Dextran

Due to their excellent biocompatibility and biodegradability, dextran and its derivatives have been extensively studied for biomedical applications such as tissue engineering and drug delivery, especially CM-dextran. CM-dextran is produced by reacting selected dextran components with an activated carboxymethyl derivative in alkali. This is a polyanionic polysaccharide with a carboxyl group content in the range of 3% - 10%.

Since its functional groups can facilitate chemical conjugation and ionic complexation with various drugs, CM-dextran has attracted the attention of scientists as a component of drug delivery systems. In addition, the hydrophilic nature of CM-dextran allows prolonged circulation time of the drug in the blood, thereby increasing the tumor-targeting efficiency of the drug. In recent years, other research groups have demonstrated that the use of CM-dextran as a hydrophilic shell can significantly improve the in vivo blood circulation and tumor-homing properties of nanoparticles. Given the excellent biological properties of CM-dextrans, the development of efficient strategies to obtain CM-dextrans and their derivatives is of great significance for advancing research in the biomedical field.

Biodegradable CM-dextran Development Services at CD BioGlyco

The insertion of carboxyl groups into dextran chains provides further opportunities to immobilize biologically active molecules of interest (drugs, enzymes, diagnostic tracers) on dextrans. The carboxyl moiety of CM-dextran can be used in a variety of reactions such as esterification, amidation with amines, Ugi or Passerini reactions. Moreover, simple ion-binding reactions can also provide a series of derivatives that bind different cationic molecules. To meet our clients' extensive scientific needs, our researchers have developed a variety of strategies to help clients develop and prepare high-quality CM-dextrans. Our services include but are not limited to:

  • Synthesis of CM-dextran

We help our clients generate CM-dextrans with good reproducibility by carboxymethylating dextran using monochloroacetic acid (MCA) in water/organic solvent mixtures under strong alkaline conditions. Moreover, we increased the degree of substitution (DS) of CM-dextran by applying optimized reaction conditions.

  • Characterization of CM-dextran

We offer a variety of techniques, such as Fourier Transform Infrared (FTIR) spectroscopy, to help clients confirm the structure of CM-dextrans.

  • Development of CM-dextran-based derivatives

We have established mature experimental protocols to help clients develop CM-dextran-based derivatives, including carboxymethyl dextran sulfate (CMDSu), carboxymethyl dextran benzylamide sulfate (CMDBSu), and carboxymethyl dextran benzylamidesulfonate sulfate (CMDBSSu).

  • Development of CM-dextran coated magnetic nanoparticles (CMD-MNPs)

We synthesized magnetic nanoparticles (MNPs) by co-precipitation and covalently bound CM-dextran to MNPs to generate CMD-MNPs, which can be functionalized and used for bioactive compound immobilization.
