Lipopolysaccharides (LPSs) are intricate molecules found in the outer membrane of gram-negative bacteria. Comprising lipid A, core oligosaccharide, and O-antigen or O-side chain polysaccharides, LPSs play a critical role in maintaining the integrity of bacterial membranes. The lipid A component, made up of fatty acids and glucosamine disaccharides, anchors LPS in the outer leaflet of the bacterial cell membrane. The core oligosaccharide contains various residues, including heptose, 3-deoxy-D-manno-oct-2-ulosonic acid, hexose, and hexosamine.

LPSs have significant implications for gram-negative bacterial biology and virulence, influencing the host immune system and its responses. They act as potent pyrogens and stimulators of the immune system. In severe cases, LPS can contribute to the development of septic shock. Moreover, accumulating evidence suggests that prolonged exposure to lower concentrations of LPS may have adverse effects on autoimmunity, obesity, depression, and cellular senescence.
