CD BioGlyco has extensive experience in Lectin Production and testing of physicochemical properties. Lectins are a class of active proteins that bind highly selectively to specific sugars. Several lectins have been isolated from G. simplicifolia seeds and leaves: GSL-I, GSL-II, GSL-III, and GSL-IV, which have different sugar-binding specificities. Currently, GSLs have been used in glycan localization, disease marker research, and so on. In order to meet the different research needs of our clients about GSL, we provide a one-stop GSL production service.
Fig.1 Process of GSL production. (CD BioGlyco)
We mainly extract GSL from the leaves and seeds of G. simplicifolia. Based on the reported methods for GSL isolation and extensive experience in Plant Lectin Production, we determine the best extraction protocol for the crude extract by optimization. The crude extract is subsequently purified by graded precipitation with ammonium sulfate. Lectins have sugar-binding specificity. Affinity columns are made by using specific sugars or glycoproteins as affinity ligands. Among the many isolation and purification methods, affinity chromatography purification of lectins is the most effective. Higher purity samples are obtained in a single purification. Therefore we use Affinity Chromatography for further purification.
The physicochemical properties of different lectins have some differences. We will complete the relevant physical and chemical property tests according to the client's requirements. We compare their sugar-binding specificity by sugar inhibition assay. We also evaluate its activity by its ability to agglutinate blood red blood cells. In addition, subunit molecular weight, etc., will also be tested.
Based on the optimized production method, we obtain high-quality and purity GSL and its conjugate products. We have a wide range of products including alkaline phosphatase conjugate, fluorescein conjugate, peroxidase conjugate, and so on.
