Guanosine 5'-diphosphate (GDP) is a nucleoside diphosphate composed of pyrophosphate unit, pentose ribose sugar, and nucleobase guanine. GDP is involved in intracellular signaling processes functioning. When an extracellular signal triggers the activation of a G protein-coupled receptor (GPCR), the associated G protein exchanges its bound GDP for GTP, activating a downstream signaling cascade, and stimulating a variety of cellular responses, containing regulated gene expression, cytoskeletal rearrangements, and modulation of enzyme activity. GTP can be hydrolyzed to GDP by the GTPase activity of the G protein, becomes inactive, and the signaling event is terminated. GDP is also a potential iron mobilizer, preventing hepcidin-ferroportin interactions. As a high-energy compound, GDP plays a vital role in cells. The mutual conversion of GDP and GTP plays a role in regulating protein activity and cell functions in cells.
